Where can I find the buffalo quest?
I dont understand what is the angle buffalo quest and what I have to do with it.

It's not "angle" but "angel", so perhaps misreading it is what has confused you.

It's in a Secret Room on the mountain, the one with 3 cows in it.

I want to find the slime king.

The slime king is completely unconnected with the buffalo quest, they are 2 separate things.
To get the quest, talk to the main in the room behind the left side mirror in the Tower.
When ready, speak to the person at the back of that room and she will take you there.
